The basics are that you must have a completed manuscript, be an unpublished author, and if you succeed through the ranks, you'll be offered a publishing contract by a Simon and Schuster imprint.
I decided to enter the contest despite the requirement to sign the contract within five days because it could act as a foot in the door should I win. As long as the contract does not tie up additional novels, it's worth it to me. If nothing else, I'll get some fun feedback from the folks who take a look and feel inclined to comment as well as rate the chapter. So far it's been great :).
Anyway, the way the contest works is people read the first chapters and "grade" them by interest. Each chapter is only posted for 14 days. You do need to join Gather.com to participate, which you can do here: http://www.gather.com/register.jsp?beamBack=viewMessage.jsp%3FmessageId%3D9851624185668385
